NAD: The Molecule Essential for Life
NAD (Nicotinamide Adenine Dinucleotide) is a molecule central to cellular function. It powers energy production and regulates DNA repair, making it crucial for maintaining life. Professor Mathias Ziegler from the University of Bergen explains:

Our cells rely on energy derived from food. NAD acts like a rechargeable battery, transferring energy from nutrients to cellular processes. This energy transfer primarily occurs in mitochondria, often referred to as the “powerhouse of the cell.”
How NAD Supports Aging and DNA Repair
As we age, our DNA accumulates damage, increasing the demand for NAD to fuel repair processes. NAD also regulates gene expression, which is essential for cellular function. However, studies reveal that NAD levels decline with age, partly due to heightened DNA repair activity.
“Decreased NAD levels are generally tolerated by cells, but prolonged depletion can impair mitochondrial function, leading to energy deficits and disease,” says Ziegler.
Ziegler’s team used advanced tools, including CRISPR-Cas9 and mass spectrometry, to uncover how mitochondria act as NAD reservoirs. These reservoirs buffer NAD levels during periods of stress, preventing cellular decline. However, excessive depletion of mitochondrial NAD can lead to dysfunction, driving aging-related diseases.
Exploring NAD Supplementation for Healthy Aging
Mitochondrial dysfunction and declining NAD levels are hallmarks of aging and age-related disorders like dementia. Clinical trials investigating NAD supplementation show promise in restoring cellular energy and slowing disease progression.
“Our findings offer new insights into the mechanisms of aging and potential therapeutic strategies,” says Ziegler.
This research, published in Nature Metabolism, highlights the importance of NAD in combating aging and opens the door to innovative treatments aimed at preserving cellular health and extending lifespan.
